Rui Therapeutics' KN5501 Initiates Phase 1 Clinical Trial for CD19-Targeted Autoimmune Diseases

KN5501: Product and Regulatory Status
KN5501 is an investigational cell therapy product, not yet branded, consisting of CD19-chimeric antigen receptor-modified NK cells derived from healthy donors, designed to eliminate CD19-positive B cells. Its off-the-shelf manufacturing approach may reduce turnaround time and manufacturing variability compared to autologous CAR-T therapies, which require patient-specific collection and production. Rui Therapeutics has reported that in a separate SLE study, four severely ill patients treated with KN5501 experienced improvement from severe to mild disease based on the SLEDAI-2K score, with related Chinese case data published in The Lancet in November 2025. However, the small sample size and lack of a randomized controlled trial mean that DLT, infection rates, cytokine release syndrome (CRS), neurotoxicity, and duration of remission will be critical determinants of investment decisions in NCT06318533. The global SLE treatment market is estimated at approximately $1.9 billion in 2024, and Benlysta's standalone sales of approximately $1.8 billion in 2025 demonstrate the unmet need and willingness to pay in severe patients.
