mRNA-LNP formulation calculator — Converts mRNA mass to phosphate moles (P = mass / 330 g/mol/nt) and computes ionizable lipid moles required for a target N:P ratio. Includes SM-102 / MC3 / ALC-0315 4-lipid system presets (Ionizable / PC / Cholesterol / PEG-lipid at 50/10/38.5/1.5 mol%) and microfluidic FRR (flow rate ratio, water:ethanol = 3:1 baseline).

How to Use

① mRNA input • mRNA mass (µg) — typical batch 100–1000 µg • Average nt MW = 330 g/mol/nt (auto) ② Target N:P ratio (typically 5–6, mRNA vaccine standard) ③ Lipid preset • SM-102 (Moderna mRNA-1273): SM-102/DSPC/Chol/PEG-DMG = 50/10/38.5/1.5 • ALC-0315 (Pfizer BNT162b2): ALC-0315/DSPC/Chol/PEG-DMG = 46.3/9.4/42.7/1.6 • MC3 (Onpattro): MC3/DSPC/Chol/PEG-DMG = 50/10/38.5/1.5 ④ Lipid MW (autofilled) ⑤ FRR (aqueous:organic ratio, typically 3:1 to 4:1) ⑥ Output: • Mass (mg) per lipid • Total lipid molar amount • Aqueous / organic flow rates (from FRR)

Examples

Example) 100 µg mRNA, N:P = 6, SM-102 preset → P moles = 100 / 330 = 0.303 µmol → Ionizable (N) = 6 × 0.303 = 1.82 µmol (SM-102) → Total lipid (Ionizable at 50 mol%) = 1.82 / 0.50 = 3.64 µmol → SM-102 (MW 710): 1.82 × 710 = 1.29 mg → DSPC (MW 790, 10 mol%): 0.364 × 790 = 0.288 mg → Cholesterol (MW 387, 38.5 mol%): 1.40 × 387 = 0.542 mg → PEG-DMG (MW 2509, 1.5 mol%): 0.055 × 2509 = 0.137 mg → FRR 3:1 → mRNA aqueous 3 mL/min + lipid ethanol 1 mL/min
