Ligation Calculator
Mass and volume arithmetic for an explicit user/source insert-to-vector molar ratio
No ratio is preselected or recommended. Enter the value and its exact user/source basis.
All component and total volumes are explicit inputs; this calculator does not supply a product recipe.
Tool Guide
Definition
Ligation calculator — From vector/insert length (bp), concentration (ng/µL), and target molar ratio, computes the insert mass (ng) and volume (µL) for a T4 ligase reaction. Formula: Insert(ng) = Vector(ng) × ratio × (Insert_bp / Vector_bp).
Purpose
(1) Check mass arithmetic for a ratio selected by an exact source profile or the user (2) Calculate DNA volume from entered concentrations (3) Preserve the ratio basis and every reaction component in the receipt (4) Never recommend a ratio, condition, or success rate
How to Use
① Enter vector and insert lengths plus one mass. ② Enter the molar ratio and its exact source/user basis. ③ Add concentrations to calculate DNA volumes. ④ Enter total, buffer, and ligase volumes from the source/user plan. Empty fields are never filled as a recipe.
Examples
Example: entering a ratio preserved in a source receipt and a vector mass derives only the insert mass. Missing ratio basis or reaction fields remain INCOMPLETE.
