fraction_cleaved = Σ(cleaved bands) / (uncut + Σ(cleaved bands))

% editing = 100 × (1 − √(1 − fraction_cleaved))

The square root correction accounts for the fact that T7E1 cleaves heteroduplexes formed between wild-type and edited alleles (Guschin et al., 2010).

  • • SpCas9 (wild-type): 20–80%
  • • High-fidelity Cas9 (eSpCas9, HiFi): 10–60%
  • • Cas12a (Cpf1): 10–50%
  • • Base editors (CBE/ABE): 10–70% (T7E1 not ideal — use sequencing)
