hERG Exposure Margin Calculator — hERG IC50・nominal steady-state total Cmax を µM に正規化し、high-exposure multiplier・plasma fu を適用して IC50/high total Cmax・IC50/high free Cmax を計算します。fu<1% の場合、ICH E14/S7B Q&A に従う計算用 fu=1% 感度値を並列表示します。
目的
(1) hERG IC50・high clinical exposure の単位を正規化 (2) Total・free Cmax margin を並列検算 (3) DDI・臓器機能低下等の high-exposure multiplier を反映 (4) IC50 lower confidence bound に基づく保守的 margin を整理
使い方
① Best-practice hERG assay の IC50・任意の lower 95% bound を入力します。 ② Nominal steady-state total Cmax、分子量、plasma unbound % を入力します。 ③ 最大治療 dose・intrinsic/extrinsic factor を反映する high-exposure multiplier を入力します。 ④ Total/free・point/lower-bound margin を同一 protocol の reference drug 分布と比較します。 単一 universal cutoff は組み込まず、本 tool 単独で TdP risk を判定しません。
